The Tight Junction Protein (TJP) is located at the topmost side of epithelial and endothelial cells. It is composed of transmembrane proteins such as claudin, occludin, JAMs, and cytoplasmic scaffold proteins like ZO-1/2/3. By connecting the actin cytoskeleton, it forms a selective barrier, separating the internal and external environments and maintaining cell polarity. Targeting TJP can repair barrier damage caused by inflammation or tumors, and is a new strategy for the intervention of enteritis, stroke, and tumor metastasis.
