SBI-477 is a chemical probe that stimulates insulin signaling by deactivating the transcription factor MondoA. It coordinately inhibits triacylglyceride (TAG) synthesis and enhances basal glucose uptake in human skeletal myocytes.

SBI-477 analog (SBI-993 analog, Cpd. 41) is a compound derived from SBI-477 that exhibits potential inhibitory effects on MondoA and/or MondoB. SBI-477 analog may reduce intracellular lipid accumulation and/or increases glucose uptake, thereby lowering blood glucose levels and/or enhancing insulin signaling, and can be used in the study of MYC-driven cancers.

SBI-993 is an analog of SBI-477 that exhibits greater potency and more favorable in vivo bioavailability and pharmacokinetic parameters than SBI-477. SBI-993 inhibits the activity of the transcription factor MondoA, thereby promoting insulin signaling. SBI-993 reduces the expression of TAG synthesis and lipogenesis genes in muscle and liver, decreases the expression of Txnip and Arrdc4, and reduces the binding of ChREBP and MondoA to promoter elements of target genes in the liver, thereby inhibiting hepatic steatosis.
