PF-DcpSi is an mRNA decapping scavenger enzyme (DcpS) inhibitor with an IC50 of 0.11 nM. When administered intraperitoneally at a dose of 30 mg kg, PF-DcpSi can improve the disease phenotype in a mouse model of spinal muscular atrophy (SMA).

Rg3039 (PF-06687859) is a potent DcpS inhibitor. DcpS is a therapeutic target for spinal muscular atrophy. Spinal muscular atrophy (SMA) is caused by deletion or mutation of both copies of the SMN1 gene which produces an essential protein known as SMN. RG3039 improves motor function in SMA mice. RG3039 also showed activity to improve survival, function and motor unit pathologies in two SMA mouse models.

JCS-1 is a potent DcpSPROTAC degrader. It non-covalently binds to DcpS via an RG3039-based warhead and recruits the E3 ligase VHL. JCS-1 facilitates the ubiquitination and degradation of DcpS at nanomolar concentrations, with a DC50 of 87 nM in MOLM-14 cells. This compound can be utilized in research on acute myeloid leukemia and other DcpS-dependent genetic disorders.
