Cysteine-linked ADCs prepared with N-aryl maleimides and incorporating the anticancer drug N-phenyl maleimide monomethyl-auristatin-E (MMAE) maintain strong cytotoxicity following prolonged serum exposure, whereas N-alkyl maleimide MMAE ADCs progressively lose potency.
