Endothelial Differentiation-Related Factor 1 (EDF1) is a 148 amino acid transcriptional coactivator that contains 1 HTH cro C1-type DNA-binding domain. It has been postulated that the protein functions as a bridging molecule that interconnects regulatory proteins and the basal transcriptional machinery, thereby modulating the transcription of genes involved in endothelial differentiation. When endothelial cells are induced to differentiate in vitro, EDF1 is downregulated, leading to inhibition of cell growth and cell polarization. EDF1 binds calmodulin thorough its IQ domain and regulates nitric oxide synthase activity through calmodulin sequestration in the cytoplasm. Though ubiquitously expressed, EDF1 is most abundant in adult liver, heart, adipose tissues, intestine and pancreas. In fetal tissues, EDF1 is most abundant in kidney. There are two isoforms of EDF1 that are produced as a result of alternative splicing events.

STK16 MPSK1 possessed DNA-binding ability and activated the TGF-beta responsive CNP promoter or vascular endothelial growth factor gene promoter which possesses a sequence element analogous to the TGF-beta responsive GC-rich element of the CNP promoter. STK16 MPSK1 did not directly activate a Smads-dependent promoter from plasminogen activator inhibitor 1 gene, but it showed enhancement in co-operation with Smad3 and Smad4. STK16 MPSK1 mRNA as well as its protein level were stimulated by TGF-beta treatment.

We found that trichostatin A (TSA), an inhibitor of HDAC (histone deacetylase), induces p16(INK4A) expression in an HBP1-dependent manner. HBP1 activates or represses the expression of some specific genes during cell growth and differentiation. HBP1 was acetylated by p3 CBP in two regions: repression domain (K297 35 37) and P domain (K171 419). HBP1 acetylation after TSA treatment was confirmed by immunoprecipitation assay. HBP1 interacted with histone acetyltransferase p3 and CREB-binding protein (CBP) and also recruited p3 CBP to p16(INK4A) promoter. HBP1 acetylation at K419 plays an important role in HBP1-induced p16(INK4A) expression.
