Members of the immunoglobulin-like transcript (ILT) family are activating and inhibitory immunoreceptors whose genes are located same locus that encodes killer cell Ig-like receptors (KIR). Leukocyte Immunoglobulin-Like Receptor Subfamily B Member 2 (LIR-2) is a type I transmembrane protein. LIR-2 is expressed primarily on monocytes and dendritic cells (DC). Human LIR-2 is produced as a 598 amino acino acid precursor including a 21 aa signal sequence, a 440 aa extracellular domain (ECD), a 21 aa transmenbrane segment, and a 116 aa cytoplasmic domain. LIR-2 binds to Classical MHCI proteins. Ligation of LIR-2 incluces Tyr phosphorylation within its cytoplasmic ITIMs, a requirement for association with SHP-1. LIR-2 mediates tolerogenic DC-induced CD4+ T cell energy in vitro and in vivo.

LILRB1, also known as CD85j and ILT2, is a transmembrane glycoprotein in the LILR immunoregulatory protein family. LILRB1 is expressed on NK cells that have expanded in response to acute HCMV infection. LILRB1 exhibits considerable diversity in the population, and polymorphisms in the LILRB1 gene have been associated with susceptibility to rheumatoid arthritis and weakly associated with HCMV disease in a subset of patients with HIV. The regulation of phagocytosis by macrophages is an additional key role of LILRB1 signaling. LILRB1 recognizes a wide variety of HLA haplotypes due to its interaction with the invariant β2M subunit of MHC class I, which suggests that this signaling axis is relevant across diverse patient populations.

LILRB4,also known as CD85k and LIR-5, ILT-3,  is an approximately 60 kDa transmembrane glycoprotein that negatively regulates immune cell activation. Mature human ILT-3 consists of a 238 amino acid (aa) extracellular domain with two Ig-like domains, a 21 aa transmembrane segment, and a 168 aa cytoplasmic domain with 3 immunoreceptor tyrosine-based inhibitory motifs (ITIM).LILRB4 is receptor for class I MHC antigens.
