Neuropilin-1 (Npn-1, previously neuropilin; also CD304) is a 130 - 140 kDa type I transmembrane (TM) glycoprotein that regulates axon guidance and angiogenesis. Two homologues, Neuropilin-1 and Neuropilin-2, are identified. Neuropilin-1 binds to semaphorin 3A, The PLGF-2 isoform of PGF, The VEGF-165 isoform of VEGF and VEGF-B. Coexpression with KDR results in increased VEGF-165 binding to KDR as well as increased chemotaxis. It may regulate VEGF-induced angiogenesis. The soluble isoform 2 binds VEGF-165 and appears to inhibit its binding to cells. NRP1 expression is regulated in EC by tumor necrosis factor-alpha, the transcription factors dHAND and Ets-1, and vascular injury. NRP1 upregulation is positively correlated with the progression of various tumors.

CD177 is polymorphic and has at least two alleles: PRV1 and NB1. Human PRV1 is a Glycosyl-Phosphatidylinositol (GPI)-linked cell surface glycoprotein that belongs to the uPAR CD59 Ly6 family of receptors. PRV1 is expressed by neutrophils and neutrophil precursors,and changes in expression serve as diagnostic markers for myeloproliferative disorders such as polycythemia vera and essential thrombocythemia. PRV1 may also be expressed by Erythroblasts, B cells, and Monocytes. NB1, a Glycosyl-Phosphatidylinositol (GPI)-linked cell surface glycoprotein, was first described in a case of neonatal alloimmune neutropenia. It is reported that CD177 functions as a novel heterophilic binding partner that engages PECAM-1 in membrane-proximal IgD6.

CD28 (Cluster of Differentiation 28) is a disulphide-bonded glycoprotein belonging to the immunoglobulin (Ig) superfamily, and structurally consists of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. Mouse CD28 is constitutively expressed on the surface of all murine T cells and on developing thymocytes as disulfide-linked homodimers or as monomers. CD28 can binds the B7-1 and B7-2 ligand, and together perform important functions in the T and B cell response pathways. B7 CD28 family members, which can augment or antagonize T-cell receptor signaling, in the regulation of central and peripheral T-cell tolerance.
