| In vitro | DDD01035881 (5 μM) selectively blocks Plasmodium falciparum microgametogenesis in a time-dependent manner. Treatment within 5 min after activation disrupts exflagellation and cytoskeletal formation. DNA replication occurs, but structural abnormalities appear[1]. DDD01035881 interferes with Pfs16 function and localization during Plasmodium falciparum microgametogenesis. DDD01035881 blocks proper vesiculation and expulsion of the parasitophorous vacuole membrane (PVM), leading to failed or abnormal egress of gametocytes[1]. |

Preparation of the In Vivo Formulation:
1) Add 100 μL of the DMSO stock solution to 400 μL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
