Apoptosis detection

Propidium Iodide (PI) is a red fluorescent dye utilized for cell staining and is suitable for fluorescence microscopy, confocal microscopy, flow cytometry, and fluorometer analysis. In aqueous solution, the Ex/Em of PI is 493/636 nm. Upon binding with nucleic acid, the Ex/Em shifts to 535/617 nm, enhancing the fluorescence signal 20-30 times.

JC-1 (CBIC2) is a fluorescent lipophilic carbonyl cyanine dye. JC-1 is used to measure mitochondrial membrane potential. When the mitochondrial membrane potential is relatively high, JC-1 converges in the matrix to form a polymer, which can produce red fluorescence (λex=585 nm, λem=590 nm). When the mitochondrial membrane potential is low, JC-1 cannot accumulate in the mitochondrial matrix and exists in the form of monomers to produce green fluorescence (λex=510 nm, λem=527 nm).

Calcein-AM (Calcein acetoxymethyl ester) is a cell-permeable green fluorescent probe commonly used for assessing eukaryotic cell viability or mitochondrial permeability transition pore, with no fluorescence itself. After hydrolysis, it emits green fluorescence with excitation/emission wavelengths: 494/515 nm.

BBR-BODIPY, a fluorescent probe, facilitates the screening of its interactions with targeted cells, induces apoptosis, and alters the expression of apoptosis-related proteins [1]. Storage instructions include protection from light.

XTT is a cell-impermeable, negatively charged tetrazolium dye that produces a water-soluble formazan when reduced at the cell surface by cellular-derived NADH and an electron mediator [1,2]. It is frequently used in colorimetric assays to measure cell proliferation, cytotoxicity, and apoptosis [3].

Z-DEVD-R110 ((Z-Asp-Glu-Val-Asp) 2 -Rhodamine 110), a rhodamine derivative featuring two four–amino acid (DEVD) peptides attached to the fluorophore [1], serves as a fluorogenic substrate for caspase-3/7. This compound is utilized for detecting apoptosis, leveraging its design to bind and indicate caspase activation.

7-Aminoactinomycin D is a fluorescent DNA stain.
