Luciferase, firefly is a bioluminescent enzyme extracted from fireflies, commonly used as a reporter gene in cellular imaging studies to quantify target gene expression levels and measure intracellular ATP content, emitting peak fluorescence at 560 nm.

Firefly Luciferase mRNA (N1-Me-Pseudo UTP) is an mRNA sequence encoding firefly luciferase, which can express luciferase protein in cells after transfection, and is commonly used in gene regulation, cell viability assay, ATP level analysis and in vivo imaging, etc. It can be used as a positive control for mRNA transfection, and yellow-green fluorescence is observed at 560 nm.

L-Luciferin acts as a competitive inhibitor of firefly luciferase and serves as an alternative substrate for light production. In comparison to D-Luciferin catalysis, increasing the concentration of the enzyme or L-Luciferin results in a reduced light output.

D-Luciferin 6'-methyl ether sodium salt functions as an inhibitor of firefly luciferase and acts as a cofactor in the Luc-catalyzed synthesis of dinucleoside polyphosphates [1] [2].
