Description
Remtolumab is a dual-variable-domain immunoglobulin with low-picomolar Kd affinity for both human TNFα and IL-17. It is used in research related to rheumatoid arthritis, psoriatic arthritis, and moderate to severe plaque psoriasis.
In vitro
Methods: Affinity and potency evaluations were performed on candidate drugs such as Remtolumab, a dual variable domain immunoglobulin (DVD-Ig™), to compare their binding capacity and biological effects on human TNF and human IL-17.
Results: As a dominant dual variable domain immunoglobulin (DVD-Ig™) candidate drug, Remtolumab stood out from other candidate drugs of the same type due to its higher affinity and potency for human TNF and human IL-17 [1].
In vivo
Methods: The neutralizing effects of Remtolumab on human TNF and IL-17 were detected in cell and mouse models, and its effects on TNF-Dgal-induced lethal effect and IL-17-induced KC production were observed.
Results: Remtolumab could functionally neutralize the activities of human TNF and IL-17, inhibit the lethal effect induced by recombinant human TNF-Dgal, and suppress the production of KC induced by recombinant human IL-17 in mouse models [1].
