Description
Azelastine hydrochloride (Astelin) is a phthalazinone derivative with antihistaminergic activity.
In vivo
In the rat basophilic leukemia RBL-2H3 cell line, Azelastine inhibited antigen (Ag)-induced TNF-alpha release (IC50=25.7 mM) and ionomycin-induced TNF-alpha release (IC50=1.66 mM). In mouse peritoneal macrophages, Azelastine suppressed inducible nitric oxide synthase (iNOS) mRNA expression and nitric oxide (NO) production. In normal human mast cells, Azelastine inhibited the secretion of IL-6, TNF-alpha, and IL-8, and the activation of NF-κB, as well as intracellular calcium levels. In human gingival fibroblasts, Azelastine dose-dependently inhibited DNA and protein synthesis.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
