Description Mezigdomide (CC-92480) is a potent, novel, selective and active cereblon E3 ubiquitin ligase modulator (CELMoD) that often acts as a molecular glue. Mezigdomide has antimyeloma activity.
In vitro Mezigdomide-induced loss of Aiolos and Ikaros in cultures of PBMCs resulted in the activation of T cells and increased production of IL-2 and IFN-γ. Mezigdomide exerts single-agent induction of apoptosis and exhibits remarkable synergy with NSC 34521. Mezigdomide is effective in CC-4047, CC-5013, and CC-220-resistant cell lines[1].
In vivo Mezigdomide (1 mg/kg) showed 75% reduction in tumor volume when tumor-bearing mice were dosed orally for 21 days. Both the 3 and 10 mg/kg doses gave near maximal response in this model[2].
