Description MI-2-2 is an inhibitor of bivalent protein-protein interaction between menin and MLL with an IC50 of 46 nM. MI-2-2 binds to menin with Kd of 22 nM.
In vitro MI-2-2 is capable of inhibiting the bivalent fragment of MLL that comprises both MBM1 and MBM2 with IC50 of 520 nM. MI-2-2 potently inhibits cell proliferation, differentiation and down-regulation of Hoxa9 expression. MI-2-2 exhibits pronounced activities at low micromolar concentrations in BMCs transformed with MLL-AF9 and in MV4[1].
