In vitro
The potent compound (SBC-115,337) demonstrated an IC50 of 0.6 μM in an in vitro ELISA assay for monitoring PCSK9 binding to recombinant LDLR. All tested compounds did not affect PCSK9 synthesis, processing, or secretion. At 1.2 μM, SBC-115,337 induced more than tenfold upregulation of LDLR in HepG2 cells, increased uptake of fluorescently labeled DiI-LDL, and lowered LDL-c levels in mice on a high-fat diet.[1]

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
