Description
Pregnenolone Carbonitrile (5-Pregnen-3β-ol-20-one-16α-carbonitrile) is an activator of rodent-PXR and induces the expression of CYP3A.
In vitro
Pregnenolone Carbonitrile is hepatoprotective by increasing resistance to subsequent stressful insults[4].
In vivo
In WT mice, Pregnenolone Carbonitrile (40 mg/kg/day; i.p.) induces the expression of CYP3A11 and CYP2B10 at the mRNA, protein, and enzymatic levels[2]. In adult female Sprague-Dawley rats, Pregnenolone Carbonitrile (100 mg/kg; i.p.) induces the expression of CYP3A mRNA[3]. In male Sprague-Dawley rats, Pregnenolone Carbonitrile (35 mg/kg; gavage) increase in Pgp expression[4].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
