In vitro
Methods: Huh7 and skHep1 cells were treated with Palbociclib hydrochloride (0.1, 0.5 μM, 10 days) to test the reversibility of cell cycle arrest.
Results: Palbociclib hydrochloride may induce irreversible cell cycle arrest in both cell lines, which may be dose-dependent. [2]
In vivo
Methods: Palbociclib hydrochloride (40 mg/kg, oral, 17 days) was used to treat NSG mice bearing IMR32 neuroblastoma cells to study its effect on tumor growth in mice.
Results: Palbociclib hydrochloride effectively inhibited tumor growth in NSG mice and had no obvious toxicity to mice. [1]

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
