Description
LY3372689 (Formulaic Ia) is an oral, highly selective, CNS-permeable O-GlcNAcase (OGA) inhibitor. LY3372689 exerts a neuroprotective effect by inhibiting OGA enzymatic activity, increasing the O-GlcNAc glycosylation level of tau protein, and thereby reducing abnormal tau phosphorylation and the formation of neurofibrillary tangles. LY3372689 can be used in research on tau-related diseases, including Alzheimer’s disease.
In vitro
Methods: 300-μm-thick mouse hippocampal slices were treated with 10 μM LY3372689 or DMSO for 4 h. Whole-cell patch-clamp recordings were performed on CA1 neurons to assess paired-pulse facilitation (PPF) and paired-pulse depression (PPD).
Results: LY3372689 significantly reduced PPF and PPD, impairing short-term synaptic plasticity. [3]
In vivo
Methods: Sprague Dawley rats were administered a single oral dose of 0.001–3 mg/kg. Plasma concentrations and enzyme occupancy rates were measured post-administration.
Results: The highest dose group treated with LY3372689 achieved over 90% EO and a maximum OGA enzyme occupancy of 98%. [1]
