| In vitro | SCFSkp2-IN-2 (Compound AAA-237) (0.3-3 μM; 24-72 h) binds directly to Skp2, leading to its proteasome-mediated degradation without affecting Skp2 mRNA expression but inhibiting Skp2 protein expression in a time-dependent manner in NSCLC cells. SCFSkp2-IN-2 inhibits the proliferation of A549 and H1299 cells and induces apoptosis in a dose- and time-dependent manner, blocks the cell cycle at the G0/G1 checkpoint by regulating the Skp2-Cip/Kip and PI3K/Akt-FOXO1 signaling pathways, and induces NSCLC cell senescence (0.3 μM; 1-7 days).[1] |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
