Description
Ganaplacide hydrochloride (KAF156 HCl) is an orally administered imidazopyrimidine antimalarial agent that inhibits Plasmodium PI4K activity. It exhibits activity against Plasmodium falciparum, Plasmodium vivax, and artemisinin-resistant parasites, including artemisinin-resistant strains, and exerts effects during both the blood and liver stages.
In vitro
Methods: The SYBR Green proliferation assay was employed to evaluate the inhibitory effect of Ganaplacide hydrochloride on laboratory-adapted Plasmodium falciparum strains and clinical dissociation strains.
Results: The IC50 values of Ganaplacide hydrochloride against multiple drug-resistant strains ranged from 6 to 17.4 nM. [1]
In vivo
Method: Ganaplacide hydrochloride (1-15 mg/kg) was administered orally for causal prophylaxis in rodent malaria model mice, and the prophylactic activity of KAF156 was monitored through blood smear analysis.
Result: A single oral dose of 10 mg/kg Ganaplacide hydrochloride administered 2 hours before infection completely protected the mice. [1]
