Description
Cilobradine hydrochloride (DK-AH 269) is an HCN Channel blocker and an open channel blocker of neuronal Ih and related cardiac If channels.
In vitro
Cilobradine hydrochloride is an HCN channel blocker that is about 3 times more potent than ZD7288. At a concentration of 10 μM, Cilobradine hydrochloride inhibits WT mHCN2 channel current by 86%. In contrast, I432A and A425G channel currents were only reduced by 14% and 19%, respectively, by this concentration of Cilobradine hydrochloride. The double mutant (I432A/A425G) channel was even less sensitive to 10 μM Cilobradine hydrochloride (8 ± 2% inhibition)[1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
