Aditoprime (Aditoprim) is a selective inhibitor of bacterial dihydrofolate reductase (DHFR) that inhibits the transformation of dihydrofolic acid to tetrahydrofolic acid. Aditoprime shows good antibacterial activity and broad antimicrobial spectrum, and moreover, excellent pharmacokinetics. Aditoprime inhibits L.casei and E.coli DHFR with IC 50 of 520 and 47 nM, respectively [1] [2] [3].

In vitro | Aditoprime exhibits an antibacterial spectrum analogous to TMP, demonstrating high susceptibility (MIC or MIC 50 ≤ 4 μg/mL) across a diverse range of bacteria. In swine, it is effective against Salmonella and Streptococcus; in chickens, against E. coli and Salmonella; in calves, it targets E. coli, Streptococcus, Mannheimia, and Pasteurella; in sheep, Streptococcus and Mannheimia; and in fish, it combats E. coli, Flavobacterium columnare, A. baumannii, and Y. ruckeri [1]. |
In vivo | Aditoprime (Aditoprim) exhibits longer elimination half-lives (3.3~14.8 h) and higher distribution volumes (4.6~10.4 L/kg) compared to Trimethoprim (TMP) across various animal species including pigs, calves, monkeys, and sheep. It demonstrates a similar pattern with its distribution volume being approximately four times greater than that of TMP. Additionally, when administered intramuscularly at dosages of 10-40 mg/kg body weight, Aditoprime effectively treats swine streptococcosis in infected swine [1]. |
