| In vitro | NGP555 effectively lowers Aβ42 in cell cultures (9 nM) and increasing shorter forms of Aβ at the same time . |
| In vivo | NGP555 also shows good oral bioavailability and is brain-penetrant with a brain: plasma ratio of ~0.93 in mice. NGP555-treated Tg mice show significant protection from a decline in performance with >65% less decline (P<0.005) when comparing the differential of Tg to non-Tg vehicle-treated mice. NGP555 obviously lowers Aβ42 in the cerebrospinal fluid (CSF) at time points from 8 to10 hours post-dose. When combining the reduction of Aβ42 with an increase in Aβ38, NGP555 is effective at raising CSF Aβ38/42 ratio at 1.87 mg/kg and above . |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
