Description
IndiMitecan (LMP776) is an inhibitor of topoisomerase I (Top1) with anticancer activities [1].
In vitro
Indimitecan (LMP776) exhibits antiproliferative effects across a variety of human cancer cell lines, demonstrating a mean-graph midpoint (MGM) for growth inhibition at 0.079 ± 0.023 μM, as per the National Cancer Institute's developmental therapeutics assay ("NCI60"). Its potent efficacy is highlighted by its ability to induce DNA cleavage through Top1 inhibition and its role as a substrate for metabolic ketone reductases. In specific cell proliferation assays involving cell lines such as HOP-62, HCT-116, SF-539, UACC-62, OVCAR-3, SN12C, DU-145, and MCF-7, Indimitecan showed growth inhibition with GI50s ranging from <0.01 to 0.08 μM, underscoring its potent antiproliferative activities across a diverse set of cancer cell lines.
