| Description | CKK-E12 is an ionizable lipid and a component of lipid nanoparticles for mRNA delivery, exhibiting organ selectivity with high targeting specificity for hepatocytes. Frequently co-assembled with other lipids to form carriers, this lipid is utilized in research and therapeutic development for RNA-related diseases. |
| In vivo | Methods: In female C57BL/6 mice, CKK-E12 lipid nanoparticles containing 2 μg mRNA were administered via intravenous injection (CKK-E12/DSPC/cholesterol/DMG-PEG2000 molar ratio 50:10:38.5:1.5). Detection was performed 6 h after single-dose administration. Results: CKK-E12 mediated mRNA expression in the liver, but its expression level was lower than that of LNPs combining CKK-E12 with optimized lipids (such as R-A11B9). [1] Methods: In female C57BL/6 mice, a single tail vein injection of CKK-E12 lipid nanoparticles was administered (containing 0.75 mg/kg luciferase mRNA or 0.5 mg/kg Cre mRNA, dissolved in citrate buffer, mixed with ethanol, and dialyzed in PBS). Results: Detection at 6 h post-administration showed that CKK-E12 primarily mediated hepatic expression; m1Ψ modification did not significantly increase overall expression but enhanced splenic expression by 8-fold. Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
