| In vivo | Methods: In female C57BL/6 mice, CKK-E12 lipid nanoparticles containing 2 μg mRNA were administered via intravenous injection (CKK-E12/DSPC/cholesterol/DMG-PEG2000 molar ratio 50:10:38.5:1.5). Detection was performed 6 h after single-dose administration. Results: CKK-E12 mediated mRNA expression in the liver, but its expression level was lower than that of LNPs combining CKK-E12 with optimized lipids (such as R-A11B9). [1] Methods: In female C57BL/6 mice, a single tail vein injection of CKK-E12 lipid nanoparticles was administered (containing 0.75 mg/kg luciferase mRNA or 0.5 mg/kg Cre mRNA, dissolved in citrate buffer, mixed with ethanol, and dialyzed in PBS). Results: Detection at 6 h post-administration showed that CKK-E12 primarily mediated hepatic expression; m1Ψ modification did not significantly increase overall expression but enhanced splenic expression by 8-fold. 2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
