| Description | Antiviral agent 34 is a potent, orally active, broad-spectrum small-molecule inhibitor against influenza A/B viruses, targeting the core site of influenza virus RNA-dependent RNA polymerase (RdRp) with an EC₅₀ of 0.8 nM against H1N1. Antiviral agent 34 can be used for studying the mechanisms of influenza virus infection and for drug development. |
| Targets & IC50 | influenza B:0.8 nM (EC50), influenza A (EC50):0.8 nM |
| In vitro | Methods: MDCK cells infected with H1N1 were treated with Antiviral agent 34 (2–125 nM) for 24 hours, followed by Western blot analysis to detect H1N1 nucleocapsid protein. Results: Antiviral agent 34 reduced H1N1 nucleocapsid protein (NP) levels in a concentration-dependent manner. [1] |
| In vivo | Methods: BALB/c mice were infected with a lethal dose of H1N1 virus, followed by oral administration of Antiviral agent 34 (100 mg/kg/day) via gavage twice daily for 7 consecutive days. Results: Antiviral agent 34 provided approximately 50% protection in infected mice. [1] |
