| Description | 2-Hydroxy-3-methylanthraquinone is a natural anthraquinone derivative that exhibits cytotoxic and pro-apoptotic activity against various tumor cells. It modulates SRC-related signaling and the activity of the IL-6/JAK2/STAT3 pathway, and induces a decrease in mitochondrial membrane potential and Caspase-3-dependent apoptosis. It can also influence MAPK-related signaling pathways, including the phosphorylation levels of p38MAPK and ERK1/2. 2-Hydroxy-3-methylanthraquinone is also commonly used in research related to anthraquinone chemistry, redox properties, and the pharmacological activity of natural products. |
| In vitro | 2-Hydroxy-3-methylanthraquinone (Compound 1) inhibits the tyrosine kinase activity of pp60c-src, GST-v-src, and SPC-A-1 total protein, with IC₅₀ values of approximately 20 μM, 33 μM, and 67 μM, respectively [1]. 2-Hydroxy-3-methylanthraquinone (48 h) inhibits the proliferation of HepG2, SPC-A-1, and Bcap37 cancer cells, with IC50 values of 51 μM, 66 μM, and 57 μM, respectively; however, its activity against SW480 cells is significantly lower (IC50 >125 μM)[1]. 2-Hydroxy-3-methylanthraquinone (10 μM; 48 h) induces apoptosis in HepG2 cells, as evidenced by nuclear chromatin condensation and a significant increase in the apoptotic cell population [1]. 2-Hydroxy-3-methylanthraquinone (2–12 h) causes a time-dependent decrease in mitochondrial membrane potential in HepG2 cells, with significant reductions observed at 6 h and 12 h [1]. 2-Hydroxy-3-methylanthraquinone (10 μM; 0–24 h) induces time-dependent activation of caspase-3 in HepG2 cells, with a significant increase in activity observed between 6 h and 24 h [1]. |
