Description
QS-21, an immunostimulatory saponin, is a potent vaccine adjuvant that stimulates both Th2 humoral and Th1 cell-mediated immune responses by interacting with antigen presenting cells (APCs) and T cells. Furthermore, QS-21 activates the NLRP3 inflammasome, leading to the release of cytokines IL-1β and IL-18, dependent on caspase-1.
In vivo
Research on mouse antigen-presenting cells (APCs), specifically dendritic cells (DCs) and macrophages, has identified QS-21 as a stimulant for the NLRP3 inflammasome. This activation leads to the subsequent release of caspase-1-dependent proinflammatory cytokines, IL-1β/IL-18, which either support the maturation of Th17 cells or energize interferon-gamma (INF-γ)-mediated Th1 responses, respectively[3].

Stock Solution Preparation:

Dissolve 2 mg of the compound in 100 μL DMSOTargetMol | reagent to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
