PDE4B-IN-2 (compound 33) does not inhibit CYP1A2, CYP3A4, CYP2C9, and CYP2D (IC50>10 µM) but inhibits LPS-induced TNF-α production in vitro in mouse peripheral blood mononuclear cells (PBMC; IC50=0.5 µM) [1].
In vivo
PDE4B-IN-2 (compound 33; 2 mg/kg; po) achieves a Cmax of 8.7 μg/mL and an AUC of 52.3 μg•h/mL in mice[1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
