Description UCB7362 (GLXC-26743) is an orally available and potent plasmepsin X (PMX) inhibitor with anti-malarial activity.UCB7362 inhibits parasite reproduction.
Targets&IC50 PMIX:142 nM, PMX:7 nM
In vitro UCB7362 exhibits significantly higher potency against PMX than PMIX, with an IC50 of 7 nM for the former compared to 142 nM for the latter[1].
Additionally, UCB7362 demonstrates improved selectivity against Cat D and Renin, with an IC50 of 3889 nM and >10,000 nM, respectively[1].
In vivo UCB7362, when administered orally at doses ranging from 10 to 60 mg/kg twice a day for 4 days, clears parasitemia from peripheral blood[1].
Furthermore, when given intravenously (IV) at 1 mg/kg and orally (PO) at 10 mg/kg in a single administration, UCB7362 exhibits moderate clearance in dogs and cynomolgus monkeys, and moderate to high clearance in rats[1].
