In vitro | Tiodazosin acts as a potent competitive antagonist of postsynaptic alpha adrenergic receptors, effectively shifting the norepinephrine concentration response curves rightward in the mesenteric artery. Analysis using a Schild plot with two Tiodazosin concentrations yields a pA2 value of 8.66 and a slope of -0.99. Furthermore, Tiodazosin blocks norepinephrine-induced contraction in the portal vein, leading to a nonparallel suppression of the norepinephrine concentration-response curve and significantly reducing the maximal response to norepinephrine. |
