Description
PARG-IN-4 (compound A) is an orally active and cell-membrane permeable PARG inhibitor (EC50=1.9 nM) that inhibits tumour growth in mice with antitumour activity.
Targets&IC50
HCC1954 cells:0.37 μM, RMUG-S cells:8 nM
In vitro
PARG-IN-4 shows strong anti-proliferative effects against ovarian and breast cancer cells. It inhibited cell growth with IC₅₀ values ranging from 8 nM (RMUG-S) to 0.37 μM (HCC1954), depending on the cell line[1].
In vivo
In a patient-derived xenograft (PDX) mouse model (HBCx-34), oral administration of PARG-IN-4 (100 mg/kg, once daily for 45 days) led to complete tumor regression, achieving a tumor growth inhibition rate (TGI) of 110%[1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
