In vitro | E235 at a concentration of 1 µM induced a time-dependent decrease in the levels of XBP-1s mRNA in HT1080 cells after 2, 4, 8, 16, or 24 hours [1]. Antiproliferative activity of E235 was observed in HT1080, RPMI-8226, B16F10, 4T1, HT1080 shNT, and HT1080 shATF4 cells when treated with concentrations ranging from 0.1 to 10 µM for either 4 or 5 days [1]. Dose-dependent upregulation of p53 expression was seen in AG1522 cells treated with 0, 1, 5, and 10 µM of E235 for 4 hours. Additionally, a 2-hour treatment with 1 µM E235 increased p-Chk2 expression in AG1522 cells [1]. Increases in p-p53 expression were eventuated in both AG1522 and HT1080 cells with E235 dosages of 0, 1, 5, and 10 µM over a 2-hour treatment [1]. Finally, E235 stimulated a dose-dependent increase in expression of γ-H2AX in HT1080 cells with treatments ranging from 0, 0.5, 1, 5, to 10 µM at a 0.5-hour interval [1]. |
