Description CGP55845 hydrochloride is a potent and selective GABAB receptor antagonist with an IC 50 of 6 nM which can be used in neurological research [1] [2].
In vitro In the mouse visual cortex, CGP 55845 hydrochloride at a concentration of 1 μM augments the frequency of both spontaneous calcium (Ca 2+) transients and inhibitory postsynaptic currents (IPSCs), including miniature IPSCs (mIPSCs), without altering the amplitude or kinetics of mIPSCs. Additionally, this compound significantly elevates the amplitudes of evoked IPSCs (eIPSCs) and reduces the paired-pulse ratio (PPR), indicating its potent effect on synaptic transmission [3].
In vivo Administering 0.5 mg/kg of CGP55845 hydrochloride intraperitoneally (i.p.; once) to Cdkl5 KO mice enhances the quantity and density of PSD95-positive puncta, alongside the development and maturation of dendritic spines within the perirhinal cortex (PRC). This treatment also reinstates novel object recognition (NOR) memory. The study utilized 2-month-old male Cdkl5 +/Y and Cdkl5 -/Y mice, observing a notable improvement in NOR memory in the Cdkl5 -/Y mice.
