| In vitro | Sperm plasma membrane integrity analysis Procedure: 1. Wash cryopreserved sperm and fresh sperm 3 times by centrifugation in PBS preheated at 36°C (3000 rpm × 5 min). 2. After washing, resuspend sperm in 1 mL PBS, mix with 10 μM PI and 1 μM 6-CFDA dye, and incubate at 37°C for 15 minutes. 3. After incubation, wash sperm 3 times with PBS and obtain images under fluorescence microscope. |
