Description
NMDI14 is a highly specific, small-molecule, non-competitive inhibitor of the nonsense-mediated RNA decay (NMD) pathway with an IC₅₀ value of 1.8 μM. NMDI14 can be used for studying NMD pathway function, PTC-related genetic disorders, and tumor research.
In vitro
Methods: Human CRC cells: DLD1, RKO (MSI-H); SW480, SW620 (MSS) were treated with NMDI14 (10 μM) for 24 h. mRNA levels of classic NMD target genes (BAG1, GAS5, ATF4) were detected by qRT-PCR.
Results: NMDI14 treatment significantly upregulated NMD target gene expression in all tested cell lines, confirming its effective inhibition of NMD. [1]
In vivo
Methods: MC38 cells were subcutaneously inoculated into C57BL/6J mice. Following successful inoculation, NMDI14 (10 mg/kg) was administered via intraperitoneal injection every other day for 23 days. Tumor volume and weight were monitored, and tumor-infiltrating immune cells were analyzed by flow cytometry.
Results: In C57BL/6J mice, NMDI14 significantly inhibited tumor growth and increased CD8⁺ T cell, DC, and NK cell infiltration along with IFN-γ levels.[1]
