| Description | NMDI14 is a highly specific, small-molecule, non-competitive inhibitor of the nonsense-mediated RNA decay (NMD) pathway with an IC₅₀ value of 1.8 μM. NMDI14 can be used for studying NMD pathway function, PTC-related genetic disorders, and tumor research. |
| In vitro | Methods: Human CRC cells: DLD1, RKO (MSI-H); SW480, SW620 (MSS) were treated with NMDI14 (10 μM) for 24 h. mRNA levels of classic NMD target genes (BAG1, GAS5, ATF4) were detected by qRT-PCR. Results: NMDI14 treatment significantly upregulated NMD target gene expression in all tested cell lines, confirming its effective inhibition of NMD. [1] |
| In vivo | Methods: MC38 cells were subcutaneously inoculated into C57BL/6J mice. Following successful inoculation, NMDI14 (10 mg/kg) was administered via intraperitoneal injection every other day for 23 days. Tumor volume and weight were monitored, and tumor-infiltrating immune cells were analyzed by flow cytometry. Results: In C57BL/6J mice, NMDI14 significantly inhibited tumor growth and increased CD8⁺ T cell, DC, and NK cell infiltration along with IFN-γ levels.[1] |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
