Description
ODN 2216 is a human-specific Toll-like receptor 9 agonist that induces robust production of interferon-α and interferon-β, activates plasmacytoid dendritic cells and conventional dendritic cells to produce IL-12. ODN 2216 indirectly stimulates IFN-γ release in peripheral blood mononuclear cells, and enhances NK cell and TCR-triggered CD4+ T-cell responses for immunological research.
In vitro
In immunological assays, co-culture of naive CD4+ T cells with ODN 2216 (6 μg/mL) for 9 days drove differentiation into Th1 effector cells via an IFN-α/IL-12-dependent mechanism. Additionally, treatment of thymic dendritic cells with 2 μM of the compound for 15 hours reduced spontaneous apoptosis rates to 23.1% compared to controls [1].
In vivo
In mouse models, ODN 2216 is used as an immunoadjuvant to elicit T-cell mediated antitumor immune responses. In liver disease research, the agonist has been employed to study the mtDNA/TLR9/MicroRNA-223 negative feedback loop. This signaling axis regulates neutrophil activation and provides protection against acetaminophen-induced hepatotoxicity in experimental animal models [3][4]

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
