Description
L-902688 is a potent, highly selective and orally active EP4 receptor agonist with a Ki value of 0.38 nM and an EC50 value of 0.6 nM. Its selectivity for the EP4 receptor is more than 4000-fold higher than that for other EP receptors and prostanoid receptors.

In vitro
Methods: Human umbilical vein endothelial cells (HUVECs) were treated with 1 μM L-902688 for 24 hours to detect its effect on the expression of Twist and α-smooth muscle actin (α-SMA) induced by TGF-β.
Results: L-902688 (1 μM, 24 h) could significantly inhibit the TGF-β-induced upregulation of Twist and α-SMA expression in HUVECs [1].
In vivo
Methods: Adult male SD rats were intraperitoneally injected with L-902688 at a dose of 0.25–1 μg/kg per day once daily for 3 weeks. The degree of right ventricular fibrosis was evaluated in a monocrotaline (MCT)-induced pulmonary arterial hypertension (PAH) model.
Results: L-902688 (0.25–1 μg/kg/day, intraperitoneal injection, once daily for 3 consecutive weeks) could attenuate right ventricular fibrosis in MCT-induced PAH rats [1].
