In vitro | IKs124 is a selective activator of KCNQ1/KCNE1 (IKs) potassium channels. In HEK293 cells expressing these channels, IKs124 enhanced IKs current in a dose-dependent manner from 0.1 to 10 μM, with maximal stimulation (~60%) observed within 5 minutes at 10 μM. IKs124 did not affect other potassium channels such as hERG or Kir2.1, demonstrating good selectivity[1]. |
In vivo | In isolated guinea pig atrial preparations, IKs124 (1 μM) significantly shortened action potential duration (APD), with more pronounced effects at higher pacing frequencies. This indicates that IKs124 enhances repolarization via IKs activation and may be useful in treating arrhythmias associated with prolonged repolarization[1]. |
