I. Solution preparation
Preparation of mother solution and working solution: 0.01-0.1 g of fluorescein sodium powder is placed in a light-proof test tube, and 10 mL of saline is added to prepare a 1-10% fluorescein sodium solution. After fully dissolving, it is used as a working solution.
II. Operation steps
Mouse kidney staining
1. The mouse is anesthetized by intraperitoneal injection of 1% sodium pentobarbital, and the kidney is completely removed during surgery.
2. Use forceps and scissors to remove the kidney surface membrane, apply fluorescein sodium to the kidney surface for 2 minutes, and then wash it three times with saline.
3. Apply 1% methylene blue staining solution to the surface for 2 minutes, wash it three times with saline after the time is up, and then observe it under a microscope at a wavelength of 470 nm.
