AMPA receptor modulator-9 is a selective, orally active positive allosteric modulator of AMPARs, capable of crossing the blood-brain barrier. It selectively enhances AMPAR activity in human and rat models, with an EC2x value of 0.96 μM in rat embryonic cortical primary neurons. The compound upregulates BDNF protein expression in primary rat cortical neuron cultures and enhances AMPA-mediated excitatory postsynaptic responses in rats and mice. | In vitro | AMPA receptor modulator-9 (Compound 14o) enhances AMPA receptor activity in primary neuronal cultures from rat embryonic cortex with an EC 2x value of 0.96 μM. In Xenopus oocytes injected with rat cortical or human hippocampal poly(AM + ) mRNA, it increases AMPA-induced inward currents (rat cortical mRNA: EC 50 = 9.9 μM; human hippocampal mRNA: EC 50 = 18 μM). Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 μL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 μL Tween 80 and mix well until fully clarified.
3) Add 450 μL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
