Bioactivity

Description	(-)-Maackiain is a natural anti-inflammatory agent that inhibits histamine H1 receptor and IL-4 gene transcription upregulation, suppresses PKCδ activation, activates the caspase-1 pathway, and promotes IL-1β production.
In vitro	(-)-Maackiain (0-30 µM, 24 hours) inhibits PMA-induced histamine H1 receptor expression in HeLa cells, while also suppressing PMA-induced phosphorylation of PKCδ at tyrosine residue 311 and preventing PKCδ translocation to the Golgi apparatus [3]. (-)-Maackiain (0-30 µM, 24 hours) suppresses DNP-HSA (antigen)-stimulated IL-4 gene expression in RBL-2H3 cells [3].
In vivo	(-)-Maackiain (5-20 mg/kg, po, once daily for 3 weeks) exhibited anti-allergic activity in a TDI-induced nasal allergy mouse model, alleviating symptoms such as sneezing, nasal mucosal swelling, nasal redness, and rhinorrhea [3].

Stock Solution Preparation:

Dissolve 2 mg of the compound in 100 μL DMSO TargetMol | reagent to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSO TargetMol | reagent stock solution to 400 μL PEG300 and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH₂O TargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
