ThioLox is a competitive 15-lipoxygenase-1 (15-LOX-1) inhibitor with an IC50 value of 12 μM, demonstrating significant anti-inflammatory and neuroprotective properties, including strong protection against glutamate toxicity.

In vitro
ThioLox (50 μM; 24 h) inhibits the expression of interleukins (IL-1β, IL-6, IL-8, IL-12b, TNFα, and iNOS) in precision-cut lung slices (PCLS). In neuronal cells, ThioLox (10 μM; 16 h) prevents mitochondrial superoxide formation and lipid peroxidation. ThioLox (5-20 μM; 14-16 h) prevents HT-22 cells from glutamate toxicity. ThioLox significantly reduces neuronal cell death[1].
In vivo
The logP value of ThioLox is 3, indicating that it is predicted to effectively penetrate the blood-brain barrier[1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
