Description
S-MGB-234 is a minor groove binder used for treating Animal African Trypanosomiasis (AAT). It shows exceptional in vitro efficacy against the main causative agents, Trypanosoma congolense and Trypanosoma vivax, without demonstrating cross-resistance with existing diamidine drugs and is not internalized through the transporters used by diamidines.
In vitro
S-MGB-234 (compound 7), with an exposure duration of 69 hours, exhibits non-toxic behavior towards L6 cells, demonstrating an effective concentration (EC 50) of 20.39 μM[1].
In vivo
S-MGB-234 (compound 7) effectively cures and prevents relapses in mice (cured/infected, 4/4) with two 50 mg/kg intraperitoneal (i.p.) doses. A partial cure (1/4) is achieved with four consecutive daily i.p. treatments of 10 mg/kg, while two 10 mg/kg i.p. doses do not cure but extend survival compared to untreated controls[1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
