Description
Ro 363 hydrochloride is an effective, highly selective agonist of β 1-adrenoceptor. Ro 363 hydrochloride is an effective inotropic stimulant. Ro 363 hydrochloride is a cardiovascular regulator, which can reduce diastolic blood pressure and significantly increase myocardial contractility.
In vitro
In isolated heart preparations from guinea-pigs, the compound Ro 363 induced arrhythmic contractions when administered at doses that elicited 70-100% of its maximal heart rate-increasing effects [1]. Additionally, in tracheal tissues of guinea-pigs that contract spontaneously, Ro 363 acted as a potent full agonist, demonstrating approximately half the effectiveness of (-)-Isoprenaline. This activity of Ro 363 is attributed to its activation of β1-receptors, as evidenced by its comparable potency to (-)-Isoprenaline in affecting the trachea, heart, and ileum [2].
In vivo
In chloralose-anesthetized felines, Ro 363 exhibits negligible arrhythmogenic activity compared to epinephrine (adrenaline), especially in subjects whose cardiac sensitization has been induced by either U-0882 or halothane [1].

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
