Product Introduction

Phenolphthalein AI Summary
Phenolphthalein exhibits a diverse range of bioactivities across various biological targets and pathways. It acts as an inhibitor of multiple thymidylate synthase enzymes, including those from Lactobacillus casei (Ki = 4700.0 nM, IF = 0.444), Pneumocystis carinii (IF = 0.608), and humans (IF = 0.182). The compound also shows cytotoxicity against mock-infected MT-4 cells (CC50 = 58000.0 nM) and inhibits the growth of several pathogens such as Staphylococcus aureus (MIC > 58000.0 nM), Streptococcus (MIC = 38000.0 nM; MGC = 58.0 uM), and Cryptococcus neoformans (MIC > 58000.0 nM). Additionally, it has shown effects on the accumulation of radioactivity in COS-7 cells and displays minimal differences in logD between pH 6.5 and 7.4, indicating a partition coefficient of 2.41. In terms of enzyme inhibition, Phenolphthalein demonstrates activity against UGT enzymes, notably UGT1A8 and UGT1A9, and has moderate action against OATP1B1 and OATP1B3 at 10 uM. It also has some effectiveness against SARS-CoV-2, inhibiting virus-induced cytotoxicity in Vero and Caco-2 cells, though with a relatively high IC50 (greater than 20,000 nM). Furthermore, it inhibits the SARS-CoV-2 3CL-Pro protease at 20µM. Despite its multiple activities, the compound shows limited oral bioavailability in humans (F ≤ 20%) and low effectiveness in inhibiting certain targets, including the HDAC6 enzyme and SARS-CoV-2 in specific cell lines. The bioactivity spectrum of Phenolphthalein indicates its potential utility as a multifaceted inhibitor with applications in antibacterial, antiviral, and enzyme inhibition therapies, though its clinical utility may be constrained by its moderate potency and bioavailability..
