Diaplasinin (PAI-749) is a highly efficient inhibitor of plasminogen activator inhibitor-1 (PAI-1) with antithrombotic activity. It can be used in cardiovascular disease research.

In vitro
Diaplasinin is an inhibitor targeting plasminogen activator inhibitor 1 (PAI-1) with an IC50 of 295 nM.[1]
Diaplasinin inhibits PAI-1 synthesis with high potency and selectivity, and maintains tissue-type plasminogen activator (tPA) and urokinase-type plasminogen activator (uPA) activity in the presence of PAI-1, with IC50s of 157 nM and 87 nM, respectively. In addition, Diaplasinin exhibits an apparent Kd value of 254 nM and an IC50 value of 140 nM by quenching the fluorescence of fluorescently labeled PAI-1. [2]

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
