| In vivo | Methods: LNPs encapsulating different siRNAs (targeting FVII [hepatocytes] or ADAMTS13 [HSCs], or control siLuc) were administered via tail vein injection at doses of 1 mg/kg siRNA (standard silencing dose) or 5 mg/kg siRNA (high dose for toxicity assessment). Animals were euthanized 1 week after dosing, and blood and liver tissue were collected for analysis. Results: Hepatocyte silencing (FVII): At a dose of 1 mg/kg, the knockdown effect of ALC-0315 LNP on plasma FVII protein levels was approximately twice that of MC3 LNP. Hepatic stellate cell silencing (ADAMTS13): at a dose of 1 mg/kg, the inhibitory effect of ALC-0315 LNP on plasma ADAMTS13 activity was approximately 10 times that of MC3 LNP. [1] Methods: Female BALB/c mice were selected and received intramuscular (IM) injections of ALC0315-DNA, ALC0315-RNA, or an empty LNP control into both tibialis anterior muscles. Imaging was performed at 6 h, 24 h, 48 h, 72 h, 7 d, 14 d, 21 d, and 28 d. Results: The signal intensity of RNA-LNP was significantly higher than that of DNA-LNP. The expression duration of ALC0315-DNA was >28 days, while that of ALC0315-RNA was 21 days.[2] |

Dissolve 2 mg of the compound in 100 μL DMSO
to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO
stock solution to 400 µL PEG300
and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O
and mix thoroughly until a homogeneous solution is obtained.
