In vitro | Pilsicainide (SUN 1165 free acid; 10-200 μg/mL) reduces the peak amplitude of the net inward current dose-dependently, exhibiting an IC50 of 29.2 μg/mL in levo-thyroxine (T4)-treated rat atrial cells [1]. |
In vivo | Pilsicainide (SUN 1165 free acid; 2 mg/kg; i.v.; once) effectively reduces the conduction velocity in the atrium of T 4-treated male Sprague-Dawley rats (weighing 200-220 g) by diminishing the Max dV/dt and the net inward current, as well as altering various electrocardiographic parameters. Administered as a bolus injection into the right external carotid vein over two minutes, pilsicainide notably extends the QT interval at both 15 and 60 minutes post-administration, decreases the durations of P wave and QRS complex, significantly reduces the action potential amplitudes (APA) and Max dV/dt, and lengthens the duration of the action potential. |
