Description
IWP L6 (Porcn Inhibitor III) is a highly potent Porcn inhibitor with an EC50 of 0.5 nM.

In vitro
In zebrafish embryos, IWP-L6 significantly inhibits the formation of posterior vertebrae, which is dependent on Wnt/β-catenin. IWP-L6 exhibits stability in human plasma (>24 hours) but shows rapid metabolism in rat plasma (half-life: 190 minutes), mouse plasma (half-life: 2 minutes), and mouse liver S9 fractions.
In vivo
In cultured embryonic kidneys, IWP-L6 can reversibly block the Wnt signaling pathway, thereby inhibiting Wnt-mediated branching morphogenesis. It also impedes the phosphorylation of Dvl2, a cytoplasmic effector of the Wnt pathway, in HEK293 cells.

Preparation of the In Vivo Formulation:

1) Add 100 μL of the DMSOTargetMol | reagent stock solution to 400 μL PEG300TargetMol | reagent and mix thoroughly until the solution becomes clear.

2) Add 50 μL Tween 80 and mix well until fully clarified.

3) Add 450 μL Saline,PBS or ddH2OTargetMol | reagent and mix thoroughly until a homogeneous solution is obtained.
